Knowledge is not knowing, IMHO.

Understanding is a form of knowledge.

I have knowledge and understanding

I choose both. You need knowledge to be able to understand and gain knowledge by understanding

I'm not sure what you mean by the question but I prefer someone who is understanding rather than one who just has knowledge.

You can have a wealth of knowledge, but not always understand how to put into practice what you know.

Does is require knowledge to have an understanding? Or is it the other way around???

Understanding.  Without it knowledge is just words.  Understanding allows you to do something with those words.
